Peptide Reconstitution Tutorial

Learn the proper technique for reconstituting lyophilized peptides with bacteriostatic water. Follow these steps carefully to ensure optimal peptide stability and effectiveness.

Gather Your Supplies
Before you begin, ensure you have all necessary materials in a clean workspace:

  • Bacteriostatic water vial
  • Sterile syringe with needle (typically 1ml or 3ml)
  • Alcohol wipes
  • Clean, flat surface

Clean the Vial Stoppers
Proper sterilization is critical to prevent contamination:

  • Wash your hands thoroughly or wear medical gloves
  • Use an alcohol wipe to clean the rubber stopper of both the peptide vial and bacteriostatic water vial
  • Allow the alcohol to air dry completely (about 30 seconds)
  • Do not touch the cleaned stoppers with your fingers

Draw Bacteriostatic Water
Carefully measure the correct amount of bacteriostatic water:

  • Remove the needle cap from your sterile syringe
  • Insert the needle into the bacteriostatic water vial
  • Invert the vial and slowly pull back the plunger to draw the desired amount (e.g., 1ml, 2ml)
  • Check the measurement markings on the syringe to ensure accuracy
  • Remove any air bubbles by gently tapping the syringe and pushing the plunger slightly

This is the most critical step - inject slowly and carefully:

  • Insert the needle into the peptide vial at a slight angle
  • Aim the water stream at the side of the glass vial, NOT directly onto the powder
  • Inject the water very slowly to avoid foaming
  • The powder should dissolve naturally as the water runs down the side
  • Do NOT shake the vial - this can damage the peptide structure

Gently Dissolve the Peptide
Allow the peptide to dissolve naturally with minimal agitation:

  • After injecting the water, gently swirl the vial in a circular motion
  • Do NOT shake vigorously - use only gentle rotation
  • The solution should become clear within 1-5 minutes
  • If powder remains, let the vial sit in the refrigerator for 10-15 minutes, then gently swirl again
  • Once fully dissolved, the solution should be clear and free of particles
Storage and Handling
Important information for maintaining peptide stability

Before Reconstitution

  • • Store lyophilized peptides in the freezer (-20°C or colder)
  • • Keep away from light and moisture
  • • Most peptides remain stable for 1-2 years when frozen
  • • Allow vial to reach room temperature before opening

After Reconstitution

  • • Store in refrigerator (2-8°C) immediately after reconstitution
  • • Most peptides remain stable for 2-4 weeks when refrigerated
  • • Protect from light (wrap in foil if necessary)
  • • Never freeze reconstituted peptides
  • • Label vial with reconstitution date and concentration
Common Mistakes to Avoid

Shaking the vial

This can denature the peptide. Use gentle swirling only.

Injecting water directly onto powder

Aim at the side of the vial to prevent foaming.

Using regular water

Always use bacteriostatic water to prevent bacterial growth.

Freezing reconstituted peptides

This can damage the peptide structure. Refrigerate only.
